UltraDx JAK2 V617F Mutation Detection Kit is used to detect the mutation of JAK2 V617F site in bone marrow and blood cells.
The gene mutation at JAK2 V617F site was detected qualitatively in vitro. The JAK family is a kind of non-receptor tyrosine protein kinases, including JAK1, JAK2, JAK3, and TYK2. Some growth factors and most cytokines can affect the transcriptional regulation of genes by JAK activating signal transducers and transcriptional activators (STAT). JAK-STAT signal transduction pathway is involved in many important biological processes such as cell proliferation, differentiation, apoptosis, and immune regulation. Some mutations in the JAK2 gene continue to activate the JAK-STAT pathway, which is the cause of tumorigenesis.
Myeloproliferative diseases (MPD) mainly include polycythemia vera (PV), essential thrombocythemia (ET), and idiopathic myelofibrosis (IMF). It has been confirmed that about 90% of PV and 50% of ET and IMF patients have JAK2 gene V617F point mutation. JAK2 gene mutation detection can be used in the diagnosis of MPD. It is now included in the diagnostic criteria for MPD by the World Health Organization (WHO).
The mutation hotspot of JAK2 V617 codon was detected by ARMS-qPCR method. Based on the basic principle of fluorescent PCR, primers and specific probes were designed to detect the targets, and the difference of the number (concentration) of the tested substances in the original samples was determined by analyzing the results of amplification.
